摘要
茶叶中含有600多种化学成分,组分极为复杂。茶叶中的无机矿物质有27种,大多数都是人体健康所必须物质。茶叶中的有机物茶多酚与儿茶素类物质、咖啡碱、茶多糖等决定了茶叶色泽、香气、滋味、营养及保健功效。本文总结了茶多酚为主要内容物的提取纯化及性质为主要内容,对比不同方法的优异,按要求选择一条合适的工艺路线。溶剂浸提法工艺简单、技术成熟。离子沉淀法选择性强、纯度较高但是损失大、收率低、安全性低。树脂吸附法虽工艺简单、纯度高、能耗低但是溶剂用量大、成本高。超临界流体萃取法纯度高但是设备要求高、成本高,不适合大剂量的提取茶多酚。超声波浸提法高效、节时、提取率高但噪音污染,不适合长期使用。微波浸提法高效、节能、节时、提取率高但具有微波辐射。膜分离法工艺简单,环境污染小但纯度低。所以根据不同的要求、设备、成本需要选择不同的方法。

1绪论
茶叶起源于我国,后流传于世界。至今,地球上引种茶树的国家已经达到了60多个,近年来世界茶叶种植面积总数达290万公顷[1]。我国是产茶大国,进入21世纪以来,我国的茶产量稳居世界第一[2]。科学研究和临床医学实验表明,茶叶有降血糖、降血脂、抗癌、抗衰老、抗辐射等诸多保健作用,这与茶叶中的有效功能成分密不可分。茶叶中的有效成分包括茶多酚、茶多糖、咖啡碱、茶氨酸、茶蛋白等。所以,用中低档茶叶为原料,提取分离有效成分,大力发展茶叶深加工技术,不仅可以开辟中低档茶叶市场、充分利用茶叶资源,也将成为我国茶叶行业发展的新方向。
茶多酚作为茶叶中最主要的功能性成分,使其成为目前天然产物研究的热点,由于具有多种生理活性和功能,在医疗保健、食品行业、日用品等领域都得到了广泛的应用。因此,优化提取工艺,分离提纯茶多酚具有十分重要的经济和社会效益。目前,国内茶多酚生产企业基本上采用的是溶剂法制备茶多酚,该方法生产周期长,温度高,所制备的茶多酚含量和活性低,且由于在生产过程中使用氯仿等有机溶剂,不仅操作不安全,产品还存在有毒溶剂残留问题,其品质难以满足国内外市场的需求,造成茶多酚产品销售困难,大量积压,同时溶剂法对环境的污染较大,不符合绿色化生产发展的方向。因此,需要选择一条合理,绿色,创新的生产工艺,提高茶多酚产品的质量,实现资源、环境、经济、社会一体化发展。
1茶叶的主要成分及功能简介
茶是我国的国饮,是天然的绿色饮料,我国是最早发现茶树,并且最早采制茶叶和饮用茶汤的国家。在我国,根据茶多酚的氧化程度,茶叶基本被分为绿茶、红茶、乌龙茶、黑茶、白茶和青茶六类[3,4],其生理活性和功能也因成分的不同有所差异。
1.1茶叶的主要成分
茶叶含有 600 多种化学成分,因而组分极其复杂,主要有无机物质和有机物质之分[5]。
1.1.1茶叶中的无机成分
只占干茶叶总质量 4%~9%的无机物质影响着人体的健康营养及茶叶的质量。茶叶中发现的无机矿物质元素(N 、H 、O 、N 除外)有 27 种:P 、B 、As 、S 、Se 、Si 、I 、F 、Br 、K 、Mg 、Mn 、Al 、Ca 、Na 、Fe 、Cu 、Ni 、Zn 、Mo 、Pb 、Cd 、Co 、Cs 、Cr 、Ti 和 V 等元素,大多数元素都是人体健康所不可缺少的[6]。
1.1.2茶叶中的有机成分
(1)茶多酚与儿茶素类物质
茶多酚是茶叶中多酚类物质及其衍生物的总称,在茶叶的药效中起主导作用。儿茶素类物质是茶多酚中最重要的一类结构复杂的化合物,儿茶素在绿茶中含量最高;红茶、乌龙茶等发酵类茶由于加工工艺的不同,儿茶素发生氧化聚合反应生成茶黄素、茶红素等有色化合物,从而对茶的品质有很大影响。
(2)茶蛋白与氨基酸
茶叶中蛋白质含量很高,达 20%~30%,但水溶性的蛋白质仅有 1%~3%。氨基酸占茶叶干重的 1%~5%,人们已在茶叶中发现 28 种氨基酸,如茶氨酸、精氨酸等。其中,茶氨酸是茶树所特有的一种氨基酸,含量占氨基酸总量的 50%以上。目前对茶蛋白和茶氨酸的生理功效进行研究表明:茶蛋白能清除超氧阴离子,具有降血脂,抗突变、抗动脉粥样硬化
,和冠心病等作用;茶氨酸具有降压,提高记忆力,增强人体抵御病毒感染的能力等作用[78]。
(3)咖啡碱
咖啡碱的含量占有机物干物质总量的 2.5%~5%,是构成茶叶滋味的重要物质;它也是一种重要的医药原料,具有强心利尿、兴奋神经中枢等作用[9-11],同时也是多种饮料的添加剂,历来为食品、医学行业人士重视。
(4)碳水化合物与茶多糖
茶叶中碳水化合物的含量占干物质总量的 20%~25%,但多数是淀粉、纤维素等不溶于水的多糖类化合物,也含有葡萄糖、果糖、麦芽糖等水溶性的游离糖。上世纪 80 年代发现茶叶中活性多糖即茶多糖有着重要的生理活性。实验表明,茶多糖具有抗血栓、抗辐射、降血脂、降血糖,增强机体的非特异性免疫能力,防治甲状腺肥大和糖尿病等多种功效[12-17]。
(5)维生素
茶叶中含有丰富的维生素,饮茶是获得人体所需维生素的极好来源。
(6)芳香类物质
茶叶中芳香类物质含量虽然不超过干物质的万分之三,但其种类却超过500多种,这些物质相互作用和配合,产生了各种独特的茶类香气[18-20]。
(7)有机酸和皂苷类物质
茶叶中含有苹果酸、金鸡钠酸等多种有机酸,有助于维持体液的平衡。皂苷类物质在茶叶中含量约为0.07%,不仅具有抗虫杀菌、抗渗消炎、抗癌等活性,而且作为一种性能优良的天然表面活性剂,广泛应用于日化、林业、纺织等行业。
(8)色素和脂质
色素占茶叶干物质总量的 1%左右,是构成茶叶色泽及茶汤颜色的成分。茶叶中脂质含量不高,约为 4%~5%,饮茶可以获得一定量的脂肪酸。
1.2茶叶的功能
《本草拾遗》记载:“诸药为各病之药,茶为万病之药”。茶叶中具有的含量丰富的营养素和多种药效成分是其独特的保健和药用作用的基础。从传说几千年前神农时代对茶的发现和利用,到现代对茶的深入研究,茶的医药和保健功能[21]不断被发现,简述如下:(1)生津止渴;(2)消暑;(3)清热;(4)提神;(5)明目;(6)解毒;(7)止痢;(8)通便;(9)降脂减肥;(10)利尿;(11)除腻助消化;(12)预防高血压;(13)防治高血脂;(14)防治脂肪肝;(15)防治糖尿病;(16)防癌抗癌;(17)补硒;(18)消炎杀菌;(19)改善心血管功能;(20)消除口臭;(21)预防龋齿;(22)防治坏血病;(23)抗辐射;(24)延缓衰老等。
2茶多酚的组成和功效
2.1茶多酚的组成及儿茶素的结构
茶多酚又名茶单宁或茶鞣质,简称 TP (tea polyphenols ),是从天然植物茶叶中提取的多羟基酚类及其衍生物的混合物。茶多酚在绿茶的含量最高,占茶叶干重的 15%~30%,因此提取制备茶多酚大都以绿茶为原料。茶多酚是由儿茶素类、黄酮醇及配糖物、无色花青素、酚酸及缩酚酸、氧化态聚合酚类等 30 多种酚类物质组成。其中儿茶素类占茶多酚总量的 65%~80%,是其最主要成分,主要包括儿茶素(catechin ,C )、表儿茶素(Epicatechin ,EC )、表儿茶素没食子酸酯(Epicatechin gallate,ECG )、儿茶素没食子酸酯(Catechin gallate,CG )、表没食子儿茶素没食子酸酯(Epigallocatechin gallate,EGCG )、没食子儿茶素没食子酸酯(Gallocatechin gallate,GCG )、表没食子儿茶素(Epigallocatechin ,EGC )、没食子儿茶素(Gallocatechin ,GC ),其中主要单体的结构式如图2 所示:

2.2茶多酚的功效
2.2.1清除自由基、抗衰老作用
茶多酚具有清除体内自由基的能力,不仅抑制机体内脂质过氧化作用或延缓细胞脂褐素的产生,而且能提高体内抗自由基损伤的酶系 SOD 和 GSH-PX 等的活性,再生体内维生素 C 和维生素E 、GSH 等高效抗氧化剂,延缓细胞衰老。

2.2.2抗辐射作用
茶多酚通过清除自由基,调节与辐射有关酶的活性,增强造血功能及杀菌等作用,有助于受损细胞的恢复,从而减小或防治辐射对人体的影响。茶多酚还能吸收 200~330nm 的紫外线,保护皮肤,被誉为“紫外线过滤器”。[24]
2.2.3抗突变作用
茶多酚可灭活终致突变剂或抑制前突变剂的代谢,抑制致癌物与 DNA 结合等,从而发挥其抗突变作用。[25-27]
2.2.4防癌和抗癌作用
它能阻断亚硝酸盐等致癌物质在体内的合成,并可直接杀伤癌细胞和增强免疫力,对肝癌、胃癌、口腔癌等多种癌症的预防和辅助治疗,均能起到良好的疗效。[28-30]
2.2.5解毒、抗过敏作用
另外,茶多酚是天然的金属离子络合剂,能使重金属离子(如 Pb 2+、Hg 2+、Cr 6+等)沉淀,并可作为生物碱和重金属盐中毒的抗解剂,起到一定的缓解作用。茶多酚对顽固性特应性皮炎中的过敏反应有良好的抑制作用,对哮喘等过敏性病
[31-33] 症亦有显著的疗效。
2.2.6消炎、抗菌、抗病毒作用
茶多酚有消炎作用,具有广谱抗菌性,其抑菌能力和选择性都很强,在一定程度上能抑制口腔细菌、螺旋杆菌、大肠杆菌、等多种病原菌的生长。儿茶素类化合物可明显抑制流感病毒,对艾滋病病毒的增殖亦可起到抑制作用。[34-36]
2.2.7降低胆固醇、降血脂作用
茶多酚通过降低血脂水平,调节血甘油三酯(TG )、胆固醇(CH )及低密度脂蛋白胆固醇(LDL-C )和高密度脂蛋白胆固醇(HDL-C )在血液中的含量,起到降低体内胆固醇和血脂的作用。[37-39]
另外,茶多酚还具有降血压和抗动脉粥样硬化、减肥轻身、护齿除口臭、降低血糖、调节机体免疫功能的功效[40-52]。
3茶多酚的提取制备工艺
3.1溶剂浸提法
溶剂浸提法是最传统的提取工艺。常用溶剂有水、乙醇、乙酸乙酯等,利用茶多酚易溶于这些溶剂的性质,使其从茶叶或浸提液中分离。工艺流程见图 3.1。

溶剂提取法是工业用于提取茶多酚最常用的方法,提取原理是茶多酚易溶于水、醇类、酮类、酯类、醚类等,可利用水和有机溶剂将茶多酚从茶叶中提取出来。常用的方法有:
①茶叶—用水或乙醇浸取—回收水或乙醇、喷雾干燥—茶多酚。
特点:工艺简单,成本低,但所生产的茶多酚粗品含有大量的多糖、色素、咖啡因、氨基酸等,茶多酚的含量仅为30%左右,儿茶素含量仅10%,市场销售价格每千克一般只几十元,应用受到限制;只有经过进一步的精制、去杂、才有实用意义。
②茶叶—用水或乙醇浸取—乙酸乙酯(丙酮) 萃取回收乙酸乙酯—喷雾干燥—茶多酚
特点:在工艺一的基础上使用乙酸乙酯作主浸取剂,或丙酮作助提剂,提高了产品的纯度和得率,且产品的抗氧化活性好,但咖啡因的含量仍较高,茶多酚含量在70%左右,市场销售价格每千克100元。
③茶叶—用水或乙醇浸提—真空浓缩—氯仿萃取—水相(含儿茶素) —乙酸乙酯萃取—回收乙酸乙酯、干燥—茶多酚
3.2离子沉淀法
离子沉淀法[53-56]是根据茶多酚能与某些金属离子络合生成沉淀物的性质,将茶多酚从浸提液中分离,实现与其他组分的初步分离。工艺流程见图3.2。

3.3树脂吸附法
树脂吸附法[57-58]是利用树脂对茶多酚的选择性吸附实现与其他组分之间的分离。该工艺需要对茶多酚选择性强的高吸附量的填料,
适用于制备少量高纯度
的茶多酚产品,不太适合工业化生产。工艺流程见图3.3。

3.4超临界流体萃取法
超临界流体萃取法[59-60]目前研究较多的体系包括 CO 2、水、氨、甲醇、乙醇等。如今人们将有毒的有机溶剂用超临界流体来代替,实现对目标产物的高效萃取分离,具体方法如下:高压下使超临界流体与茶叶接触,控制体系的温度和压力使茶多酚溶解在超临界流体中,进而升温或降压降低其密度,使茶多酚析出。该法由于其介质多是无毒的 CO 2,尤其适合医药、食品等行业中产品的制备。工艺流程见图3.4。

3.5超声波浸提法
超声波浸提法[61]是先利用超声波的机械破碎和空化作用使细胞破碎,加速茶多酚等浸提物从茶叶向溶剂的扩散速率,缩短浸提时间,再采用与传统工艺相同的制备过程从浸提液中取得产品。传统工艺的浸提时间都比较长,而且在高温下还容易造成茶多酚的氧化,使得率降低;而超声浸提时间短,避免了茶多酚长时间处在高温环境下的氧化,使产品得率和质量都有所提高。
3.6微波提取法
微波浸提法[62-64]是利用分子在微波场中发生高频运动的特点,加速茶多酚等浸提物的扩散,进而使其快速浸取出来。该法省时、高效、节能,可有效地保护有效成分。Pan Xuejun 等人[65]使用微波辅助的方法提取茶多酚和咖啡碱,提取效率和茶多酚得率都有所提高。
3.7膜分离法
膜分离法[66-67]是将浸提液流过半透膜,以膜两侧产生浓度梯度为传质推动力,使溶液中茶多酚等小分子可以选择性地通过膜,一定程度上将茶多酚、咖啡碱等与蛋白质、果胶、可溶性淀粉等进行了分离。该方法工艺简单,条件温和,环境污染小,但是制备的茶多酚纯度不高,仍需要进一步纯化。
4结论
提取天然产物的方法根据其性质不同,方法会所改变。提取茶多酚根据其溶解性,酸性,与金属离子络合反应等性质确定了多种不同的提取分离茶多酚的方法。表 1中总结了茶多酚提取工艺的优缺点,根据提取的不同要求选择不同的提取方法。
