巨大芽孢杆菌发酵培养基的优化

摘要:为了得到巨大芽孢杆菌(Bacillus megaterium)发酵的最佳培养基,在单因子试验的基础上,应用Plackett-Burman设计法对影响芽孢产量的基本培养基组分中的关键因子进行了筛选,并进一步采用响应面分析法(RSM)对影响芽孢产量的关键因素最佳水平作了深入的研究。结果表明,影响巨大芽孢杆菌芽孢产量的关键因素是豆粕和淀粉的浓度。通过RSM的拟合和推算得到在豆粕和淀粉浓度分别为48.85和19.67 g/L时,此时模型预测发酵最佳产量为43.16×108 CFU/mL,验证值为42.2×108 CFU/mL,预测值与验证值之间吻合较好,比原始培养基提高了约10倍。

Abstract: To obtain the optimal medium for producing Bacillus megaterium spore, statistical method was used to investigate the medium components for spore concentration and productivity by Bacillus megaterium. The Plackett-Burman design was used to evaluate the effects of six variables. The results showed that the optimal values of the two parameters determined by RSM were soya bean meal of 48.85 g/L and starch of 19.67 g/L. The spore production was predicted to be 43.16×108
CFU/mL, and the actual value was 42.2×108 CFU/mL. They were fitted well with 10 folds increase. Key words: Bacillus megaterium; Plackett-Burman; response surface analysis methodology (RSM)
